Pharmacological effects: Ivermectin has a good killing effect on nematodes in the body and arthropods on the body surface. The mechanism of its deworming effect is to promote the release of gamma aminobutyric acid (GABA) from presynaptic neurons, thereby opening up GABA mediated chloride
Ionic channel. Ivermectin also exhibits selectivity and high affinity towards glutamate mediated chloride ion channels located near GABA mediated sites in invertebrate neurons and muscle cells, thereby interfering with signal transmission between neurons and muscles, causing the parasite to relax and paralyze, leading to death or expulsion from the body. The inhibitory interneurons and excitatory motor neurons of nematodes are their sites of action, while the site of action of arthropods is the neuromuscular junction. The expulsion rate of adult and fourth stage larvae of sheep's blood lance nematode, Oster nematode, Cooper nematode, hairy roundworm (including Eisenia's hairy roundworm), roundworm, upside down nematode, slender neck nematode, hairy head nematode, esophageal mouth nematode, net tail nematode, and sheep's Charcot nematode is 97% to 100%. It is also effective for arthropods such as maggots, mites, and lice. The therapeutic effect on chewing lice and sheep ticks and flies is slightly inferior. Ivermectin is also highly effective against ticks and flies breeding in feces. Although the drug cannot immediately kill ticks, it can affect feeding, molting, and egg laying, thereby reducing reproductive capacity. The effect on blood flies is similar. The elimination rate of roundworms, red pig roundworms, orchid roundworms, hairy head nematodes, esophageal mouth nematodes, posterior roundworms, toothed tail nematodes and immature worms in pigs is 94% to 100%. It is also highly effective against intestinal trichinella (ineffective against muscular trichinella) and has a good control effect on pig blood lice and pig scabies mites. It is ineffective against flukes and tapeworms.
The pharmacokinetics of ivermectin vary significantly depending on the animal species, dosage form, and administration route. The bioavailability of subcutaneous injection is higher than that of oral administration, but oral administration absorbs more quickly than subcutaneous injection. After absorption, it can be well distributed to most tissues, but it is not easy to enter the cerebrospinal fluid. The apparent distribution volumes of sheep and pigs are 4.6 L/kg and 4 L/kg, respectively. Most animals have a longer half-life, with sheep and pigs having a half-life of 2-7 days and 0.5 days, respectively. This product undergoes metabolism in the liver, mainly hydroxylation in sheep, and methylation in pigs. Mainly excreted from feces, less than 5% is excreted in its original form or metabolic products from urine. Up to 5% of the administered dose is excreted from the milk of lactating female livestock.
Drug interactions: Simultaneous use with ethamine may result in severe or fatal encephalopathy.
